Development of classical chromatography methods like ion exchange chromatography or hydrophobic interaction chromatography for protein purification require substantial effort, time and budget. In order to reduce the number of experiments, we want to set-up a model capable of predicting protein elution and therefore separation performance. An important part of model set-up is the determination as well as a solid understanding of the interstitial, column and pore porosity. Parameters such as column packing, flow rate, particle size distribution of the beads or wall effects, among others, can affect the porosity of a column.
The student accepted for the project/thesis described above will on the one hand experimentally measure porosities on anion exchange resins and on the other hand do an intense literature research and evaluation on particle packing models and theory.
The student successfully applying for this project/thesis is skilled in written and spoken English to familiarize her/himself with the current and relevant literature of the topic described above and to fluently communicate within the international atmosphere at Fraunhofer IME. (S)He has a good basic knowledge in biochemistry and bioengineering and is willing to gain more insights into the issues faced by today’s pharmaceutical biotechnology. Experience in downstream processing as well as data handling and presentation in Excel and PowerPoint will greatly facilitate the progress and success of the successful applicant.
